The present invention relates to an optimizing method of manufacturing a solid starter using Saccharomyces cerevisiae Y204 (KACC93237P). Specifically, as a result of verifying an optimum condition for manufacturing a solid starter of Saccharomyces cerevisiae Y204 (KACC93237P), the following findings were confirmed: the growth rate and the number of live bacteria were the highest when a solid starter of S. cerevisiae Y204 is cultured on a YPD culture medium at a stirring speed of 150 rpm at a temperature of 25 °C for 24 hours; and a survival rate of a starter is increased when 10% of skim milk, 1% of lactose, and 7-10% of trehalose are added respectively when a sold starter is manufactured. Additionally, it is verified that freeze-drying showed 1.2-1.4 times fewer live bacteria but resulted in a contamination rate that was approximately twice as low compared to drying with hot air when the solid starter of S. cerevisiae Y204 enzymes is dried. Particularly, in producing the solid starter of the strain, the following findings were confirmed: enzyme activity of the strain was high when 10% of a liquid starter was inoculated and cultivated for six days at 40°C; the number of spores of the solid starter was high when 5% of the liquid starter was inoculated and cultivated for five days at 40°C; and the number of bacteria in the solid starter was lower, the lower the amount of liquid strain inoculation and the longer the fermentation period. Therefore, the production method can be useful in producing the solid starter of Aspergillus luchuensis 74-5 (KACC93235P) having low bacteria count.

The present invention relates to a method for preparing solid culture starter for preparing unrefined rice wine by using Aspergillus oryzae 75-2(KACC93236P) being indigenous strains having excellent saccharification performance and the solid culture starter for preparing the unrefined rice wine prepared by using the same. More specifically, strains for the solid culture starter are confirmed to have high enzyme activities when inoculating 5% of liquid starter strains and culturing the solid culture starter for 6 days at 35°C. The strains for the solid culture starter are confirmed to have a higher number of seeds in the solid culture starter when inoculating 10% of liquid starter strains and culturing the solid culture starter for 6 days at 40°C.
