... 5-(Hydroxymethyl)furfural(5-HMF)은 주로 식품을 굽는 과정에서주로 생성되며, 향미를 증가시키는 물질로 알려져 있다. 과거의 연구에서는 잠제적 위험을 가질 것으로 추정하였으나, 최근에는 5-HMF의 항산화, 항염증 등 다양한 효과가 보고되고 있다. 이번 발명에서는 5-HMF의 장관면역에 대한 효과와 기전를 in-vitro와 in-vivo 실험을 통하여 확인하였다. Caco-2 세포을 이용한 5-HMF의 in-vitro 에서는 연접단백질인 오클루딘과 클라우딘1의 발현량의 증가와 p-ERK의 발현량의 감소가 농도 의존적으로 나타났다. 5-HMF의 in-vivo 실험에서 5-HMF의 투여가 장투과성을 현저하게 낮추는 효과를 확인하였다. 그리고 마우스 혈액에서 대표적인염증성사이토카인인 IL-6와 TNF-a의 발현을 측정한 결과도 5-HMF의 투여량 의존적으로 감소하였다. 또한 마우스의 장조직에서 western blot을 통해 측정한 COX2와 pERK의 발현도 5-HMF의 투여량에 따라 감소하는 것을 확인하였으며, 마우스의 장조직의 H&E 염색을 통한 조직병리학적 분석에서도 융모손상과 궤양의 정도도 5-HMF의 투여량 의존적으로 감소하였다. 이러한 실시예의 결과를 종합해 볼 때, 5-HMF가 마우스의 장관에서 염증을 완화하고, 장관벽을 강화시켰다. 이러한 결과를 바탕으로 5-HMF는 장관 면역개선과 위, 장관 염증 예방 및 억제효과를 가지는 치료용 약학 조성물 및 건강기능식품, 보조사료 사료첨가제 조성물로 활용할 것을 기대할 수 있다.
