Provided is an application of a sulfhydryl single-chain DNA in the specific amplification of a reinforced polymerase chain reaction. A proper amount of sulfhydryl single-chain DNA is added to a PCR system to realize PCR amplification, the proper amount indicating that the final concentration of sulfhydryl single-chain DNA in a 20 μL reaction system is not less than 15 μM. The sulfhydryl single-chain DNA (1) is an arbitrary sequence not complementary with the target sequence; (2) Tm≥37.7°C; and (3) at least one end comprises a sulfhydryl alkyl group SH-C6H12-.
